Microplate preparation:
NGM was prepared following standard procedure. NGM variable volume (3,4 and 5ml) was dispensed to 6 well microplate (GBO brand with lid) to obtain 4.5mm, 5.5mm and 6.5mm agar width. One hour later, 20ul OP50 were added to each well and cultured overnight at room temperature.
Experiment:
30 C.elegans, young adult N2 strain, were transferred to each well of a microplate 6well format containing NGM variable volume. Technical duplicates were employed.
Locomotor activity of worms was recorded twice using WMicrotracker ARENA system for 150minutes, at 25ºC. 2nd acquisition was performed turning 180º the microplate. Acquired data was analyzed using MSExcel.
Results:
The system is able to detect locomotor activity in all NGM volume conditions, Non statistical differences has been observed employing 3ml , 4ml or 5ml of medium.
